Rose was born Ernestine Potowska in Piotrków Trybunalski, Poland, in 1810. Her father was a rabbi, and her mother was a devout Catholic. Rose was raised in a traditional Jewish home, but she rejected the religion of her parents at a young age. She was also a strong advocate for the rights of women.

In 1836, Rose married William Rose, a Polish-born American journalist. The couple moved to the United States in 1839, and they settled in New York City. Rose quickly became involved in the women's rights movement. She was a founding member of the National Woman Suffrage Association, and she served as the organization's president from 1869 to 1870.
